brings together leading scientists in medicine (dermatology, pediatrics, internal medicine and surgery), molecular biology, immunology, biomedicine, and systems research to investigate fundamental mechanisms underlying development of diseases related to desmosome disorders.
Within the scope of this SFB/TRR several projects of the Department of Vegetative Anatomy of the LMU in Munich were approved. One of the projects A03 – “Super-resolution force spectroscopy to delineate early pathogenic effects of autoantibodies and cytokines in desmosome dysfunction” will be carried out here and led by Michael Fuchs.
Project no A03
In this project we will investigate the biophysical binding properties of desmosomal cadherins and the precise sequence of signaling events following a pathogenic stimulus, i.e. autoantibodies or cytokines. For this purpose, atomic force microscopy (AFM), single molecule force spectroscopy (SMFS), and stimulated emission depletion (STED) microscopy will be used in combination with biochemical approaches. The main goal is to decipher the precise spatiotemporal sequence of events following a pathologic stimulus as well as the interplay between steric hindrance and altered signaling pathways.
